Ectomycorrhizal Fungal Diversity on Abies korea and Taxus cuspidata at Two Altitudes in Mt. Halla

Abstract

In this study, the community structures of the ectomycorrhizal (ECM) fungi in the roots of Abies koreana and Taxus cuspidata were investigated at different altitudes of Mt. Halla. We identified the collected ECM root tips based on morphological characteristics and phylogenetic analysis through sequencing of the rDNA ITS regions. From the roots of A. koreana and T. cuspidata, 11 species and 12 species were identified, respectively. The Shannon’s index and species evenness and abundance of the ECM fungi were higher in the higher than lower regions, regardless of host plant species, however, the number of ECM root tips showed the opposite pattern. The community similarity among the ECM fungi in A. koreana was significantly higher than that among the ECM fungi in T. cuspidata or than that between A. koreana and T. cuspidata (p < 0.05). These results could be useful for the conservation and management of the habitat of A. koreana, which is threatened with extinction due to increasing ambient air temperature.

서론

기후는 생물다양성 패턴을 결정하는 주요인이며[1, 2], 기후의 변화는 동식물의 지리적 분포를 저위도에서 고위도로, 저지대에서 고지대로 변화시켰고[3], 서식지 이동에 실패한 종들은 멸종하기도 한다[4]. 이러한 기후 변화와 생물다양성에 관한 많은 정보들은 기후 변화로 인한 생물 종과 군집의 변화를 예측할 수 있게 한다[5]. 기후와 생물 다양성과의 상관 관계를 연구할 때 위도에 따른 생물 분포는 기후 요소 외에도 시간적, 지리적 요인들이 복합적으로 작용하기 때문에 고도에 따른 생물 분포를 주로 연구한다[6]. 지구 온난화에 따른 기온 상승은 고산 지역에서 고도별 기온분포대의 상승을 야기시켰고 생물 서식 환경의 변화로 서식지 이동을 초래하였다. 고산 및 아고산 지대에 서식하는 수종은 산 정상부에 고립되어 있어 새로운 서식지로 이동이 어렵고 저지대에서 올라오는 수종과의 경쟁으로 개체군 감소나 멸종 위험에 처해 있다.

구상나무(Abies koreana E.H.Wilson)는 한라산을 포함하여 가야산, 금원산, 덕유산, 백운산, 속리산, 영축산, 지리산 등 고산 및 아고산에 한정되어 분포하는 우리나라 특산 식물로[7, 8] 기후변화에 취약한 종으로 평가되고 있으며[9], 최근 기후변화로 개체 군이 축소되어 세계자연보전연 맹(IUCN)에서 멸종위기종(Endangered species)으로 지정되었다[10]. 구상나무의 모든 서식지에서 생장쇠퇴가 나타나고 있는데, 기존 연구들은 그 원인을 지구온난화로 보고 있다. 기온 상승은 고산 및 아고산과 같은 한랭다습한 기후에 적응한 생물종들에게 악영향을 미친다. 주목(Taxus cuspidata Siebold et Zucc.)은 중국 북동지역, 일본, 한반도에 분포하며, 우리나라에서는 남쪽 한라산에서부터 북쪽 승적산에 이르는 해발 700~2,500 m 범위의 아고산 지대에 서식하는 상록침엽교목이다[11]. 세계자연보전연맹(IUCN)에서 관심대상종(Least concern)으로 기후변화에 취약한 종은 아니나, 덕유산과 한라산 지역에서는 고사 개체가 증가하고 있다[11].

외생균근균(Ectomycorrhizal fungi, ECM)은 산림 생태계의 주요 구성 요소로 식물 뿌리에 공생하며 숙주 식물의 영양분 흡수 및 화학물질이나 병원체, 수분 부족으로 인한 환경 스트레스를 개선하는 것으로 알려져 있다[12]. 고도 및 위도에 따른 생물 다양성은 주로 거대 동식물에서 연구되어 왔으며, 미생물에서의 연구는 저조한 편이다. 고도가 증가함에 따라 토양 미생물이 감소한다는 연구 결과가 있었으며[13], 미생물 분류군에 따라 고도별로 차이가 나타난다는 연구 결과도 있다[14]. 외생균근균의 종 다양성 및 군집은 지형과 기후 요인에 의해 변화될 것으로 예상되어 왔으며[15], 숙주 특이성[16], 종 간 경쟁 및 상호작용[17], 토양 환경[18], 대기 조성[19]등 다양한 생물적, 비생물적 요인에 의해 영향을 받는다고 알려져 있다. 온대림에서 열대림으로 갈수록 외생균근균의 종 다양성이 감소한다는 연구결과가 있지만[20] 중간 고도나 낮은 고도에서 오히려 높게 나타난다는 보고도 있으므로[21] 고도에 따른 외생균근균의 다양성을 일반화하기는 어렵다.

구상나무는 지구 온난화와 관련하여 크게 주목받고 있으며, 구상나무 보전 및 서식지 복원을 위한 많은 연구들이 진행되고 있다. 특히 구상나무 쇠퇴의 원인 중 하나로 겨울 기온의 상승으로 수목은 광합성을 할 수 있는 조건이 충족되나, 광합성에 필요한 수분공급이 부족하여 수분수지의 불균형이 발생하기 때문이라는 추정이 있다[22, 23]. 또한 구상나무와 동일한 소나무과에 속하는 소나무의 고사 원인으로 균근 생성 이전 시기의 고온 및 가뭄 발생으로 인해 균근 활동이 부재하기 때문이라는 연구 결과가 있다[24].

본 연구는 기후변화로 인해 멸종 위기에 처한 구상나무와 상대적으로 기후변화의 영향을 적게 받은 주목의 외생균근균 군집을 비교하기 위한 기초 연구의 일환으로 서로 다른 고도에서 서식하는 구상나무와 주목 뿌리의 외생균근균 군집 및 다양성을 확인하였다. 향후 이 연구를 토대로 외생균근균을 활용한 구상나무의 현지 내 보전(in-situ conservation)과 관리를 위한 기초 자료를 마련하고자 한다.

재료 및 방법

외생균근균과 토양 채집 및 형태학적 동정

연구에 사용된 기주 식물 뿌리 및 근권 토양의 수집은 2018년 6월, 제주도 한라산 해발고도 1,510-1,560 m의 하부 지역(N33°37', E126°53')과 1,860-1,910 m의 상부 지역(N33°36', E126°53')에서 수행되었다. 관음사 등산로를 따라 올라가며 구상나무림이 시작되는 부분을 하부, 끝나는 부분을 상부로 구분하였다. 구상나무와 같은 소나무과의 식물을 선정하고자 하였으나, 채집지에 서식하지않아 구상나무 주변에 함께 서식하는 주목을 선정하였다. 하부와 상부 지역에서 각각 구상나무와 주목의 뿌리 및 근권 토양을 수집하였다. 고도에 따라 5개체의 구상나무와 주목의 뿌리를 채취하였으며, 수종 간의 상관 효과를 최소화하기 위해 10 m 이상 간격을 두고 채집을 실시하였다[25]. 채취한 뿌리는 실험실로 가져와 체에 받쳐 증류수로 여러 번 씻어 뿌리에 남은 흙을 제거하였다. 세척이 끝난 각각의 뿌리를 petri-dish에 넣어 해부현미경을 사용하여 뿌리의 외생균근의 형 태학적 분류 특성에 따라 구별하였다[26]. 외생균근의 색상, 분지형태, 질감, 균사의 형태 등을 이용하여 서로 다른 형태형으로 분류한 후, 각 형태형 별로 외생균근의 수(number of ectomycorrhizal root tips)를 측정하였고, 분자 분석을 위해 균투를 냉동 보관하여 DNA 분석에 이용하였다.

외생균근균의 분자생물학적 동정

형태형에 따라 분류한 후, 냉동 보관한 균투는 외부의 이물질을 제거하기 위해 멸균수로 여러 번 세척한 후, DNeasy Plant mini kit (Qiagen, USA)를 이용하여 genomic DNA를 추출하였다. 외생균근균의 ribosomal DNA의 ITS (internal transcribed spacer) 영역을 증폭하기 위해 균 특이 프라이머인 ITS1F (5'-CTTGGTCATTTAGAGGAAGTAA-3')와 ITS4 (5'-TCCTCCGCTTATTGATATGC-3')를 이용하였고, annealing온도를 50℃로 설정하여 polymerase chain reaction (PCR)을 수행하였다[27]. PCR이 완료된 후 1.5% agarose gel에서 20분간 전기영동하여 대략 550 bp 크기의 밴드를 확인하고 DNA 염기서열 분석을 의뢰하였다(SolGent, Korea). 분석된 염기서열은 미국 국립생물정보센터(NCBI, http://ncbi.nlm.nih.gov)에서 BLAST하여 가장 일치도가 높은 종을 확인하였으며, MEGA7.0 프로그램을 이용하여[28] 계통수를 작성하고 이를 바탕으로 종을 동정하였다. 형태적 특징과 분자 분석 결과를 종합하여 외생균근균의 종 풍부도(relative abundance)를 구하고, 샘플 별로 외생균근균의 종 다양성 지수(Shannon􏌿s diversity index), 종 균등도(species evenness)를 구하였다. 또한 PCA (principal component analysis) 분석 방법을 이용하여 군집 구조를 분석하였다.

결과 및 고찰

한라산에서 채집한 구상나무 뿌리에서 분리한 외생균근균을 형태적, 분자적으로 동정한 결과 하부에서 6속 8종, 상부에서 7속 10종의 서로 다른 외생균근균을 확인하였고 상, 하부에서 모두 우점종은 Clavulina cinerea였다(Table 1, Fig.1). 선행 연구들에서 Clavulina cinerea는 내륙 산림 생태계에서 2-3번째로 우점하는 종으로 알려져 있고[29], 우리나라에서는 동해 연안의 잣나무 숲에서 높은 빈도로 나타났다는 연구 결과가 있다[30].

주목 뿌리에서는 하부에서 4속 8종, 상부에서 6속 10종의 외생균근균이 확인되었고, 하부와 상부 모두에서 Tomentella stuposa가 우점하였다. 숙주 식물에 따라 유의미한 차이를 보이는 외생균근균은 없었으며, 숙주 식물과 고도에 상관없이 모든 샘플에서 공통으로 나타난 종은 Tomentella stuposa였다(Table 1, Fig.2). Tomentella 속의 균들은 침엽수 및 활엽수림에서 흔히 발견되는 종으로[31, 32], 겉씨 식물[25]과 속씨 식물[33] 뿐만 아니라 난초[34]와도 공생하는 외생균근균으로 알려져 있다. 균주 별로 살펴보면, Russula favreiSebacina dimitica는 구상나무에서만 발견되었으며,하부와 비교했을 때 상부에서 유의미하게 높게 나타났다. Russula 속의 종들은 온대 산림 생태계에서 흔하게 나타나는 외생균근균이며[35], Sebacinaceae는 온대, 열대[36], 북극[37]과 고산 지역[38]에서도 외생균근성 숙주식물과 공생하며 상호작용하는 것으로 알려져 있다.

숙주 식물과 고도가 외생균근균 군집 구조에 미치는 영향을 비교하기 위해, Two-way ANOVA를 실시한 결과, 종 다양성지수(Shannon􏌿s index), 종 균등도(species evenness), 종 수, 외생균근(ectomycorrhizal root tip) 수에서 유의미한 차이는 없었다. 하지만 숙주식물과 관계없이 하부에 비해 상부에서 종 다양성 지수(p=0.052), 종 균등도(p=0.015), 종 수가 높게 나타났으며, 외생균근 수는 오히려 하부에서 높게 나타났다. 고도가 증가함에 따라 외생균근균의 종 풍부도가 감소한다는 결과가 있지만[39], 중간 고도나 낮은 고도에서 오히려 높게 나타난다는 보고도 있다[21]. 또한 본 연구와 비슷한 300 m 고도 범위에서 Pinus sylvestris의 뿌리에 공생하는 외생균근균의 군집을 분석한 연구에서는 고도에 따라 종 풍부도에는 차이가 없었지만, 외생균근균 군집 구조에는 차이가 나타났다[40]. 한라산에서 구상나무는 제주조릿대의 영향으로 상층에 비해 중하층에서 우점도가 현저히 감소하고 있는데[41], 이는 하부에서 외생균근균의 종 다양성 감소 원인으로 작용했을 것으로 생각된다. 실제 하부에서 제주조릿대의 밀도가 매우 높았으며, 숙주식물 뿌리와 제주조릿대 근경이 뒤엉켜 있었다. 이렇게 다른 식물과의 경쟁이 심각한 환경에서 숙주식물과 더 강한 특이성을 보이는 외생균근균 중 일부가 높은 빈도로 우점하여 종 다양성 지수를 감소시키는 것으로 보인다. 주목 역시 하부에서 제주조릿대와의 경쟁이 심해 Tomentella stuposa와 같은 일부 종과의 강한 우점 현상이 종다양성지수 감소 요인으로 생각된다. 반면 상부는 구상나무와 주목 모두 제주조릿대와 경쟁이 심하지 않지만, 기후 변화로 인해 다른 고지대 수목에 비하여 고사목 발생 빈도가 높고, 고도가 증가함에 따라 고사율도 증가한다. 특히 구상나무 쇠퇴의 주원인이 건조한 동절기 기후로 인한 수분스트레스[42] 및 그로 인한 수분수지 불균형[22] 등인 점을 고려하면, 수분 스트레스에 반응하여 숙주 식물에게 수분을 공급하는 외생균근균과의 안정적인 공생관계는 온난화로 인한 부정적인 영향을 완화시킬 수 있어 하부에 비해 외생균근균과의 상호작용이 더 활발하고 이는 종 다양성 증가로 이어졌다고 생각된다.

Principal component analysis (PCA) 군집 분석 결과 채집 고도 보다는 숙주식물에 따라 외생균근균 군집의 유사도가 높게 나타났으며, 구상나무의 경우는 고도에 따라 외생균근균 군집에 차이가 있었다(Fig. 3). 구상나무 외생균근균 군집과 주목 외생균근균 군집의 군집 간 유사도 분석을 실시한 결과, 구상나무 뿌리에서 발견된 외생균근균 군집 간의 유사도는 주목 뿌리의 외생균근균 군집보다 높게 나타났다(Fig. 4). 상부에서 종 다양성 및 종 균등도, 종 수 등은 숙주식물 간에 유의미한 차이를 보이지 않았으나, 숙주 식물에 따른 군집 내 유사도 지수가 군집 간 유사도 dimitica는 모든 군집에서 나타났다. 반면에 주목 뿌리의 외생균근균 군집은 모든 종의 상대 수도와 상대 빈도가 대체적으로 낮았다. 이는 숙주 식물에 따라 외생균근균의 군집 구조가 영향을 받으며, 구상나무 뿌리에서 발견되는 외생균근균 군집이 주목 뿌리에서 발견되는 외생균근균 군집에 비해 특이적인 군집 구조를 가진다는 것을 의미한다.

혹독한 고산 환경에서 서식하는 구상나무와 같은 고산 식물은 외생균근균과의 공생 관계에 크게 의존한다[43]. 구상나무는 기온 조건이 매우 중요하며, 자생지의 강한 일사량과, 바람, 낮은 온도 때문에 수분 스트레스에 매우 민감하다. 이러한 상황에서 구상나무의 생존과 생장에 중요한 역할을 하는 외생균근균의 확인과 군집 구조는 구상나무의 증식과 자생지 관리에 중요한 단서를 제공할 것이다. 또한 고도 변화에 따른 토양 수분과 온도 변화와 같이 잠재적인 환경 요인들에 대한 추가적인 자료 수집과 분석을 통해 기후 변화와 구상나무의 생장, 고사율 및 외생균근균의 군집 변화 등에 대한 다각적인 후속 연구가 이루어져야 한다.

한라산 상부와 하부에 서식하는 구상나무와 주목의 뿌리에서 각각 외생균근균의 군집 구조를 분석하였다. 형태적 특징과 분자적 분석을 통해 구상나무에서는 하부에서 6속 8종, 상부에서 7속 10종의 외생균근균을 확인하였다. 주목에서는 하부에서 4속 8종, 상부에서 6속 10종의 외생균근균을 확인하였다. 외생균근균의 종 다양성 지수, 종 균등도, 종 수는 숙주식물과 관계없이 상부에서 높게 나타난 반면, 균투 수는 하부에서 높게 나타났다. 하부와 상부에서 구상나무와 주목 뿌리의 외생균근균 군집 간의 유사도를 분석한 결과, 고도에 관계없이 구상나무의 뿌리에서 발견된 외생균근균 군집 간의 유사도가 주목 뿌리의 외생균근균 군집 간 유사도나 구상나무 뿌리의 외생균근균 군집과 주목의 뿌리 외생균근균 군집 간의 유사도에 비해 더 높게 나타났다. 이는 구상나무가 주목과는 다른 특이한 외생균근균 군집 구조를 이루고 있으며, 기온 상승으로 멸종 위기에 처한 구상나무의 보전과 자생지 관리에 도움을 줄 수 있을 것으로 판단된다.
