Characteristics of Cellulolytic Enzymes of Bacillus licheniformis CY-24 Isolated from Button Mushroom Compost

Abstract

The present study was performed to improve the technique used for fermenting the mushroom growth medium. Taxonomic analysis of 16S rDNA sequence from the predominant Bacillus strain CY-24 isolated during the fermentation phase of the rice straw medium identified it as Bacillus licheniformis. In addition, the growth environment of B. licheniformis was also examined in this study, which revealed the optimal growth temperature and pH to be 30 ℃ and 6.0, respectively. This study also revealed that carboxymethyl cellulase (CMCase) and polygalacturonase (PGase) enzymes isolated from B. licheniformis achieved their maximal activities at 50 ℃ and 60 ℃ respectively. Furthermore, the study confirmed that the two enzymes, i.e., CMCase and PGase in B. licheniformis are stable at temperatures above 60 ℃. The present study thus demonstrates that B. licheniformis CY-24 possesses excellent enzymatic properties. It also reveals that the action of enzymes during the production of growth mediums used for the cultivation of mushrooms is closely associated with the promotion of fermentation and softening of the rice straw. Overall, this study provides elementary information regarding the role of B. licheniformis enzymes during growth medium fermentation for Agaricus bisporus cultivation.

서론

양송이는 분류학상으로 담자균문(Basidiomycota), 주름버섯목(Agaricales), 주름버섯속(Agaricus), 양송이종(A. bisporus)에 속하는 식용버섯으로 맛과 향기가 뛰어나서 세계적으로 가장 널리 소비되는 버섯 중 하나이다[1]. 양송이 재배 방법은 지역과 나라마다 차이가 있으나 우리나라에서는 1960년대 초 부터 볏짚을 이용한 양송이 재배법의 개발로 재배를 시작하였다[2]. 볏짚은 식물 세포벽의 기본 구성성분인 cellulose 15-40%, hemicellulose 30-40%, lignin 20%, pectin 20%로 견고한 조직으로 이루어져 있으며[3], 볏짚의 세포벽을 분해하기 위해서는 배지 발효 단계에서 질소 성분이 많은 계분을 처리하여 미생물이 보다 쉽게 볏짚을 분해하도록 한다[3,4]. 버섯은 cellulase, xylanase, laccase 등 목질섬유소 분해효소를 생산하여 분해 산물인 당을 에너지원으로 이용하며, 양송이 균사는 생장 동안에 hemicellulose, cellulose, lignin 순으로 사용한다[5,6]. Cellulase는 복합효소로서 endoglucanase, exoglucanase 및 β-glucosidase 등 3가지 효소들로 구성되어 있으며, 세 가지 효소가 동시에 존재할 때 상승작용에 의하여 결정형 섬유소가 효과적으로 분해될 수 있는데 기질의 활성도 특성에 따라 carboxymethyl cellulase (CMCase), Avicelase, p-Nitrophenyl-β-D-glucoside (PNPGase)로 구분하기도 한다[7,8].

양송이 배지 생산에 있어 가장 중요한 요인은 배지의 균일성을 확보하는 것이며, 미생물의 작용에 의해 cellulase와 hemicellulase의 분비로 볏짚 배지의 C/N율 낮아지고 유기물 대비 무기물의 함량이 높아진다[4,9]. Cellulase를 생산하는 곰팡이로는 Trichoderma viride, T. koningii, Crysposporium pruinosum, Penicillium funiculosum, P. iricesis, Fusarium solani 등이 있으며[10-14], 세균으로는 Bacillus sp., Pseudomonas sp., Clostridium sp. 등이 알려져 있다[15-18]. 곰팡이는 산성과 중온에서 강한 활성을 보이는 반면 세균의 경우 내열성 또는 중성, 알칼리성에서 활성을 나타내는 균주들이 많다[7]. 특히 Bacillus sp. 유래의 미생물들은 분비 효소인 cellulase, xylanase, laccase 등을 분비하여 난분해성 물질을 분해하고 탄소원으로 효과적으로 이용할 수 있으며[19-21], surfactin, fengycin, bacillomycin과 같은 항진균성 항생물질을 생산하므로 여러 식물병 방제 등 유용한 균주로 사용 가능하다[22-24]. 또한 Bacillus sp.는 액체배양 시 생육 속도가 빠르고 포자를 형성하여 고온에서도 생육할 수 있어 퇴비화 부숙 촉진 미생물이나 미생물 제재로 개발 가능성이 높은 자원으로 많은 연구에 활용되어지고 있다[25,26,27]. 2017년 Lee의 양송이 관행 배지 발효와 밀접한 관련이 있는 미생물의 밀도 변화 연구 결과에 따르면 세균, 방선균, 형광성 Pseudomonas spp., 사상균 등 다양한 미생물들이 분포한 것을 확인한 바 있다[2,4,6].

본 연구는 양송이 배지 발효 단계 중 2차 뒤집기 과정에서 분리한 우점균을 단일 분리하여 16S rDNA 염기서열을 근거로 미생물을 동정하였으며, 발효 단계에 중요하게 작용하는 미생물의 섬유소 용해 효소(cellulase, hemicellulase, protease, pectinase) 활성을 조사하여 분리 미생물이 분비하는 CMCase, Polygalacturonase (PGase)의 효소학적 특성에 대해 보고 하고자 한다.

재료 및 방법

미생물 분리

충남 부여 석성면 농가에서 양송이 퇴비배지 2차 뒤집기 단계에서 배지를 상, 중, 하 3군데로 나누어 시료를 채취하였다. 미생물의 분리는 멸균 증류수를 이용하여 희석하고 표준평판법으로 R2A [28] (yeast extract 0.5 g, proteose peptone no. 3 0.5 g, casamino acid 0.5 g, dextrose 0.5 g, soluble starch 0.5 g, dipotassium phosphate 0.3 g, magnesium sulfate heptahydrate 0.05 g, sodium pyruvate 0.3 g, agar 15 g) 배지에 배양하였다. 배양 후 우점균 단일 colony를 독립적으로 분리하여 배양하였다. 순수분리한 미생물은 R2A 배지에서 2일 동안 배양한 후 균체를 모아 20% glycerol (V/V)에 보존하여 -70℃ 심온동결고에서 보존하면서 시료로 사용하였다.

분리 미생물 동정

분리 미생물의 동정을 위하여 API 50 CHL carbohydrate test kit (BioMerieux, Craponne, France)를 사용하여 nutrient broth (NB) 배지에 24시간 배양하였고, 배양액의 색도변화 결과를 ATB identificationcomputer system (BioMerieux, Craponne, France)을 이용하여 동정하였다. 그 후에 16S rDNA 유전자 분석을 실시하였다. DNA는 Qiagen genomic DNA lsolation kit (Qiagen, Germantown, MD, USA)을 사용하여 분리하였고, PCR 증폭은 Techne thermocycler (Bibby Scientific Limited, Stone, UK)로 수행하였다. PCR 반응 혼합액은 PCR buffer (10 mM Tris-HCl pH 9.0, 50 mM KCl, 2.5 mM MgCl2, 0.01% gelatin and 0.1% Triton X-100), 2.5 mM dNTP, 0.6 Unit Tap polymerase (Sigma-aldrich, St. Louis, MO, USA) 10 pmol fD1 (5’-AGAGTTTGATCCTGGCTCAG-3’)와 rP2 (5’-ACGGCTACCTTGTTACGACTT-3’) 그리고 50 ng template DNA로 이루어졌다. PCR은 94℃에서 1분, 56℃에서 1분 그리고 72℃에서 2분간 30 cycles로 수행하였고, 반응 후 High pure PCR product purification kit (Bioneer, Daejeon, Korea)를 사용하여 정제한 후 Genotech (Daejeon, Korea)사에 의뢰하여 염기서열을 분석하였다. 종 유사성 결정을 위해 clustal W 분석프로그램을 이용하여 데이터를 가공한 뒤 GenBank에 있는 다른 염기서열과 비교하였으며, Neighbor-joining 방법으로[29] 계통수를 작성하였으며, 계통수의 각 그룹에 대한 신뢰도는 bootstrap 1,000반복하여 분석하였다.

분리 미생물의 조효소액 조제

실험에 사용된 조효소액은 minimum salt (0.5% yeast extract, 0.5% poly peptone, 0.5% NaCl, 0.02% MgSO4·7H2, 0.1% K2HPO4) 효소 생산 배지에서 분리 미생물을 30℃에서 36시간 동안 진탕 배양한 후, 원심분리를 이용하여 배양액을 3,000 rpm에서 10분간 실시하고 상등액과 균체를 분리하여 상등액을 동결건조한 후 -20℃에 보관하면서 실험에 사용하였다.

분리 미생물의 생육조건 및 효소활성 측정

분리 미생물의 배양 적온을 조사하기 위하여 R2A 액체배지에 균주를 접종한 후 4℃에서 70℃까지 10℃ 간격으로 조정된 진탕배양기에서 180 rpm으로 24시간 배양한 후 UV spectrophotometer를 이용하여 흡광도 600 nm에서 생육 정도를 조사하였다. pH에 따른 생육정도는 pH 3.0에서 pH 9.0로 조정한 후 미생물을 접종하여 조사하였다. 19종의 효소활성을 API-ZYM kit (BioMerieux, Craponne, France)를 사용하여 Bergey’s Manual of Systematic Bacteriology [30]에 준하여 실험하였다.

미생물 효소 생산용 배지 조제

양송이 배지 제조 과정 중 볏짚 내 미생물의 효소 생산용 배지는 minimum salt (MS)배지를 기본 배지로 사용하였으며, cellulase와 polygalacturonase 활성은 기본 배지에 1.0% carboxymethyl cellulose (CMC) (pH 7.0)와 0.5% polygalacturonic acid (pH 5.0)를 첨가한 액체 및 고체 배지를 사용하였다. 그리고 hemicellulase는 기본 MS 배지에 0.5% xylan을 첨가하였고, protease는 0.5% skin milk을 첨가하여 사용하였다. 균의 배양은 10℃에서 70℃ 까지 10℃ 단위로 설정하였고, pH는 3.0-9.0에서 150 rpm으로 36시간 동안 진탕배양 하였고 균주의 생육 정도는 UV spectrophotometer를 이용하여 600 nm의 흡광도로 측정하였다.

세포벽 분해효소 활성 분석

분리 미생물의 세포벽 분해효소(cellulase, hemicellulase, protease, pectinase)활성을 조사하기 위하여 한천 확산법(agar diffusion method)을 사용하였고, 효소활성의 정도는 효소생성용 고체배지에서 생성된 clear zone의 직경을 측정하여 효소활성 정도를 조사하였다.

섬유소 분해 효소인 CMCase 활성은 CMC를 기질로 하여 측정하였다. 1.0% (w/v) CMC 용액 0.5 mL와 200 mM sodium phosphate buffer (pH 7.0) 0.25 mL를 조효소 용액 0.25 mL와 혼합하여 50℃에서 15분간 반응시킨 후 유리되는 환원당을 DNS 용액 (3,5-dinitrosalicylic acid 7.5 g, NaOH 14.0 g, sodium potassium tartrate 216.1 g, Na2S2O5 5.9 g, phenol 5.4 mL/L) 3mL을 가하고 100℃에 5분간 반응 후 상온에서 식혀 530 nm에서 흡광도로 측정하였다.

펙틴분해효소인 p-Nitrophenyl-β-D-glucoside (PGase) 활성은 polygalacturonic acid를 기질로 하여 측정하였다. 0.5% (w/v) polygalacturonic acid (PGA, sodium salt)용액 0.5 mL과 100 mM sodium acetate (NaOAC) buffer (pH 5.0) 0.3 mL의 반응 혼합액 0.8 mL를 40℃에서 미리 10분간 방치 후 0.2 mL의 조효소액을 첨가하여 40℃에서 20분간 반응시킨 후 유리되는 환원당을 DNS법으로[31] 가하고, 100℃에 5분간 반응 후 상온에서 식혀 530 nm에서 흡광도로 측정하였다.

최적 온도 및 최적 pH 설정에 따른 활성도 평가

효소의 최적 온도를 조사하기 위하여 효소 반응액 0.5 mL과 조효소액 0.25 mL을 혼합하여 4, 20, 30, 40, 50, 60, 70℃에서 20분간 반응시켜 유리되는 환원당을 DNS법으로 조사하였다. 최적 pH의 조사를 위하여 50 mM citrate-phosphate buffer (pH 3.0, 4.0, 5.0), 50 mM sodium phosphate buffer (pH 6.0, 7.0, 8.0), 50 mM glycine-NaOH buffer (pH 9.0)를 사용하였고, 완충액 0.5 mL에 효소 반응액 0.25 mL과 조효소 용액 0.25 mL을 혼합하여 20분간 반응시킨 후 DNS법으로 상대적인 활성을 조사하였다. 온도에 대한 안정성은 효소 반응액에 조효소액을 첨가하여 4, 20, 30, 40, 50, 60, 70℃에서 24시간 동안 보관한 후 효소활성을 조사하였고, pH에 대한 안정성은 최적 반응 pH 조사에 사용된 완충액 pH 3.0-9.0에 조효소액을 1 mL에 넣고 24시간 보관 후 DNS법으로 상대적인 활성을 조사하였다.

금속이온 영향 분석

금속이온은 효소 활성에 있어 촉매제 또는 저해제로 사용될 수 있으므로, CMCase와 PCase에 대한 금속이온과 상관관계를 조사하기 위해 효소 반응액에 CaCl2, MnCl2, CoCl2, BaCl2, CuCl2, MgCl2 6종 금속이온을 1 mM과 5 mM의 농도가 되도록 하여 20분간 반응시킨 후 유리되는 환원당을 DNS법으로 조사하였다. 대조구로는 10 mM EDTA를 사용하였다.

통계처리

실험은 모두 3회 반복 수행하였으며 얻어진 결과의 통계처리는 SAS program (Statistical analytical system V9.2, SAS Institute Inc., Cary, NC, USA)을 이용하여 평균과 표준편차 및 Duncan multiple range test를 통하여 각 실험 평균차에 대한 통계적 유의성 검정을 p<0.05 수준에서 항목들 간의 유의적 차이를 검정하였다.

결과 및 고찰

분리 미생물의 동정

먼저 분리 미생물 CY-24 (accession no. KT989579) 균주의 생화학적 특성을 검토하여 Table 1에 나타내었다. 분리한 CY-24 균주의 생화학적 특성은 B. licheniformis는 glycerol, glycogen, L-arabinose, D-ribose, D-xylose, D-galactose, D-glucose, D-fructose, D-mannose에서 양성반응을 나타냈으며, gelatin을 액화시키는 특징을 가지고 있었다. API-ZYM kit를 이용한 가수분해 효소활성을 측정한 결과(Table 2) esterase, leucine arylamidase, acid phosphatase, β-glucosidase 처리군에서 양성반응을 나타내었으며, 이외 효소에서는 활성을 나타내지 않았다. 이러한 결과를 바탕으로 동정한 결과 Bacillus 속(genus)으로 추정되었으며, 16S rDNA의 PCR 증폭에 의해 약 1.3 kb의 염기서열을 확보하였으며, 이 염기서열을 Ribosomal database project를 이용하여 표준균주와 상동성을 비교 분석한 그 결과 CY-24 균주는 Bacillus licheniformis와 99% 신뢰도를 보였다(Fig. 1). 동정한 결과를 토대로 분리 균주를 Bacillus licheniformis CY-24로 명명하였다. B. licheniformis는 그람 양성, 포자 형성 세균으로 환경에 부양 균으로 널리 분포되어 있으며, B. subtilis 와도 가까운 근연관계를 형성 하고있다[32,33]. B. licheniformis는 고온성이며 혐기성 세균으로 내생포자로 토양에 남아있으며, cellulose를 분해하는데 사용되는 endo (1,4) β-d-glucanase를 생산한다. Exo (1,4) β-d-glucanase는 β (1,4) glycosidic 연결을 무작위로 절단하면서 cellulose 중합체의 환원 또는 비환원 말단에서 절단되어 cellobiose 단위로 생성된다. cellobiose는 이당류 포도당으로 가수분해될 수 있으며 cellulose가 풍부한 물질을 효소적으로 가수분해할 수 있다[34]. 또한 B. licheniformis는 인간에게 비병원성 세균으로 알려져 있으며, 항균 단백질 생산과 같은 상업적 용도 및 생물학적 방제제 등 농업적 용도로 산업 효소 제조에 사용되어온 미생물로 알려져있다[35-37].

분리균의 온도 및 초기 pH의 영향

분리 미생물의 최적 생육온도를 규명하기 위하여 다양한 온도를 설정한 결과를 Fig. 2에 나타내었다. B. licheniformis CY-24 균주의 최적 생육온도는 30℃였으며, 40℃까지 생육을 하였으나, 60℃ 이상에서는 생육을 하지 못하였다. pH에 따른 생육을 조사한 결과 최적 pH는 4.0이고, pH 6.0 까지 완만하게 자라는 것을 확인하였다. Gomaa [38] 연구 결과에 따르면 30℃가 최적 온도 였고, pH는 6.0이 최적이라고 보고하였고, Vijayalakshmi [39]는 최적 온도는 40℃, 최적 pH가 6.0이라고 보고한 결과와 유사하였다. B. licheniformis는 높은 온도에서 생육이 가능하고 또한 생육의 범위가 넓은 것으로 판단된다.

분리균의 효소학적 특성

분리한 B. licheniformis CY-24 균주의 온도와 pH 변화에 따른 CMCase, PGase 효소의 활성을 조사한 결과를 Fig. 3에 나타내었다. B. licheniformis CY-24 균주의 CMCase에 대한 활성은 30℃에서 급격히 활성이 높아지기 시작하여 60℃에서 최대 활성을 보였으며, PGase에 대한 활성은 4℃ 낮은 온도에서 부터 점차적으로 상승하여 60℃에서 최대 활성을 보였으며 70℃ 지속되는 것을 확인하였다. 이 결과는 양송이 볏짚 배지의 초기발효와 후발효를 거치면서 55-65℃ 까지 온도가 상승함에 따라 미생물의 활성 또한 안정적으로 활성화 될 것이라고 생각된다. Lee 등[40]의 연구에 따르면 Bacillus 속에 속하는 균주는 45℃ 및 55℃에서 효소활성이 안정하게 유지되며, 65℃에서는 시간이 경과와 함께 효소활성이 감소한다고 보고한 결과와 비슷한 결과를 얻을 수 있었다. 또한 pH의 경우 CMCase과 PGase 두 효소 모두 pH 6.0 까지 점차적으로 상승하다 pH 7.0 부터 완만하게 낮아지는 공통점이 있었으며, PGase의 경우 강산성인 pH 3.0 에서도 80%의 활성을 나타내 CMCase는 보다 넓은 범위에서 높은 활성을 나타내는 것을 확인하였다.

금속이온 별 효소 활성

금속이온은 효소활성에 있어 촉매제 또는 저해제로 사용할 수 있으므로[38], CMCase와 PGase에 대한 금속이온 영향을 살펴보기 위하여 BaCl2, CaCl2, CoCl2, CuCl2, MgCl2, MnCl2, 6종의 금속이온을 첨가하여 효소활성을 조사한 결과를 Fig. 4에 나타내었다. B. licheniformis CY-24 균주의 CMCase의 금속이온 활성 경우 1 mM 및 5 mM의 CoCl2에서 가장 높은 활성을 보였다. 금속이온 6종 모두 60% 이상의 높은 활성을 나타냈으며, 대체로 5 mM의 농도에서의 금속이온은 1 mM 농도 보다 활성이 감소하였지만 5 mM MnCl2 농도에서는 1 mM MnCl2 농도보다 활성을 증가시켰다. PGase의 경우 6종의 금속이온 모두 80% 이상의 높은 활성을 보였으며, 이 중 1 mM CoCl2, 5 mM MgCl2의 농도에서 가장 높은 활성을 나타내었다.

적 요

본 연구는 양송이 배지의 관행 발효기술을 개선하고자 실험을 실시하였다. 양송이 볏짚 배지 발효단계에서 우점하는 Bacillus strain CY-24 균주를 순수분리하여 16S rDNA 염기서열을 통한 동정 결과 Bacillus licheniformis로 밝혀졌다. B. licheniformis 최적 생육 온도는 30℃, 최적 생육 pH는 6.0에서 가장 생육이 활발한 것을 확인하였으며, B. licheniformis CY-24 균주는 glycerol, glycogen, L-arabinose, D-ribose, D-xylose, D-galactose, D-glucose, D-fructose, D-mannose 등의 탄소원을 잘 활용하였고, 효소활성 측정 결과 esterase, leucine arylamidase, acid phosphatase, β-glucosidase 등에서 양성반응을 나타내었다. CMCase 활성 온도는 50℃, PGase는 60℃에서 가장 효과가 극대화되었으며, CMCase, PGase 두 효소는 60℃ 이상에서도 안정성이 유지되는 것을 확인하였다. 금속이온의 촉매 효과는 CoCl2 1mM에서 가장 활성이 높은 것으로 나타났다. B. licheniformis CY-24의 효소학적 특성은 매우 우수하였으며, 양송이 재배에 사용되는 배지를 생산하는 과정에 효소의 작용은 볏짚의 발효 촉진과 연화 작용에 밀접한 관계가 있으므로 유용생물자원으로써 발효기술에 대한 기초자료로 이용될 수 있을 것으로 판단된다. 추후 다양한 미생물과 버섯균의 상호작용에 대한 면밀한 연구가 이루어진다면 유용한 자원으로 이용 될 수 있을 것으로 생각된다.
