Comparison of Mycelial Growth Characteristics According to Culture Conditions of Ulleungdo Collection Strains

Abstract

The collection of biological data of indigenous species must comply with the Nagoya Protocol. Fungi contain various bioactive substances making them an attractive source of several products, including food and medicines. In this study, we investigated the growth characteristics of five indigenous fungal strains, Fomitiporia punctata, Polyporus ulleungus, P. brumalis, Gymnopus subnudus, and Tyromyces kmetii, isolated from samples collected in the Ulleungdo Island. The growth rates for each strain were assessed across various temperatures (20 ℃ to 35 ℃), culture media (Potato dextrose agar, Malt extract & Yeast extract agar, Malt extract agar, Malt extract & peptone agar, Sabouraud dextrose agar, and Modified Melin-Norkrans agar), and pH conditions (4.0 to 8.0). Additionally, we assessed the mycelial growth characteristics in liquid culture. The mycelial growth in different media varied across species; specifically, F. punctata (in MMNA), G subnudus (in MMNA), and P. brumalis (in MEPA) showed rapid growth. Optimal growth temperatures ranged between 25 ℃ and 30 ℃ for most species, with the exception of T. kmetii and P. brumalis, which were able to grow across all the temperatures tested. P. brumalis showed the best growth rate, whereas P. ulleungus showed the lowest growth potential. The optimal pH conditions for mycelial growth ranged between 4.0 and 5.0. In experiments using culture flasks, the dry weight of the culture filtrates decreased with the increasing incubation time and showed a significant decrease between 1 and 6 months of incubation, indicating that the five strains take longer than a month to fully use the culture media. Our findings highlight and establish the optimal growth conditions for five different fungal species that can be used in future application studies.

서론

버섯은 분류학적으로 담자균(Basidiomycota)과 일부 자낭균(Ascomycota) 중 일반적으로 자실체(fruiting body)를 형성하며 육안으로 관찰 가능한 고등균류를 지칭한다[1]. 버섯은 수분이 많으며 지질 함량이 적고, 단백질과 식이섬유, 무기질 비타민 등이 풍부해 오래 전부터 식품 및 약용으로 널리 이용되어 왔으며, 분석 기술의 발전으로 항산화, 항암, 항염증, 면역증강 등 다양한 약리적 효능이 밝혀지고 있다[2-5].

이처럼 버섯의 다양한 생리활성을 새롭게 밝히고 활용하기 위해서는 기존에 연구가 미진한 종 혹은 신종이나 미기록종과 같은 새로운 연구대상을 찾는 노력이 필요하다. 또한 최근 세계적으로 유전자원 접근 및 이익공유(access to genetic resources and benefit-sharing, ABS)에 대한 법령이 시행됨에 따라, 자국내에 자생하고 있는 생물자원을 확보하고 이용 가능한 자원의 특성에 대한 기초자료를 수집하는 일이 더욱 중요하게 여겨지고 있다[6]. 새로운 종을 발굴하기 위해서는 다양한 환경에 대한 조사가 수반된다. 특히 국내에서 제주도와 더불어 전형적인 해양성 기후를 보이는 울릉도는 육지와는 다르게 독립된 독특한 생태환경을 지니며 연평균 기온 12.4 ℃, 평균 강수량 1586 mm, 연평균 강수 일수 195일, 평균 습도 74%의 기후조건을 갖고 있다. 이와 같은 높은 습도와 긴 강수 일수는 버섯 생육에 최적의 조건이라고 할 수 있다[7]. 또한 면적에 비해 다양한 식물이 서식하고 있어 식물과 공생관계를 이루는 균류의 다양성도 높을 것으로 예상된다[8-9].

울릉도 서식 버섯의 종 다양성 연구를 위해 2년간(2015-2016) 채집조사를 진행하였다. 그 결과 울릉도 나리 분지(盆地) 산림지역, 성인봉지역, 석포 둘레길 그리고 내수전 둘레길 주변 산림지역 등지에서 13목, 54과, 105속 213종에 속하는 약 330여점의 표본을 확보하였으며, Tyromyces kmetii 등을 포함한 13종의 한반도 미기록종 및 Polyporus ulleungus를 포함한 2종의 신종을 찾아 보고한 바 있다[10-15]. 채집된 표본으로부터 분리배양을 통해 균주를 확보하였으며, 이 중 신종/미기록종 균주에 대한 생육조건 등의 연구를 통해 향후 생리활성 연구를 위한 생육정보를 얻고자 연구를 진행하였다. 본 연구에 사용된 대상 균주는 신종/미기록종에서 분리된 5종(Fomitiporia punctata, Polyporus ulleungus, Gymnopus subnudus, Tyromyces kmetii, Polyporus brumalis)이며, 해당 균주는 국립생물자원관 국가생물자원 배양센터에 기탁하였다.

본 연구를 통해 울릉도에서 분리된 신종 및 미기록종 균주에 대한 기초 생육정보를 얻고자 배양 조건(조성, 온도, pH)에 따른 균사 생장력, 액체배양을 통한 균사체의 생장 특성 관찰, 배지조성과 중∙단기 배양기간에 따른 배양여액 건조량의 변화를 측정함으로써 충분한 배양기간에 대한 고찰을 진행하고자 하였다[16]. 배양여액의 건조중량 비교는 균주의 생육에 따른 배양배지의 이용률을 확인하여, 향후 대량배양 조건 수립시에 참고자료로 활용하고자 하였다.

재료 및 방법

실험 대상 균주

본 연구에 이용된 균주는 울릉도에서 2015-2016년에 채집된 자생종인 Fomitiporia punctata(편절층구멍버섯), Polyporus ulleungus (울릉구멍장이버섯), Polyporus brumalis (겨울구멍장이버섯), Gymnopus subnudus (담갈색꽃애기버섯), Tyromyces kmetii 등 5종의 자실체 조직으로부터 분리 배양된 균주이며, 균주들은 국립생물자원관(National Institute of Biological Resources, NIBR)의 국가생물자원 배양센터(생물소재은행)에 기탁 보존하였다(Fig. 1; Table 1). 실험 진행전에 균주들의 종 정보에 대한 재확인을 위해 AccuPrep Genomic DNA extraction kit (K-3032, Bioneer, Daejeon, Korea)를 이용하여 genomic DNA를 추출하였다. 추출된 gDNA를 template으로하여 중합효소 연쇄반응(PCR)으로 Internal transcribed spacer (ITS) region을 증폭한 후, 염기서열분석은 마크로젠(Macrogen, Seoul, Korea)에 의뢰하였다[17-19]. 분석결과로 얻은 염기서열은 미국 NCBI의 Nucleotide BLAST (http://www.ncbi.nlm.nih.gov/blast)를 통해 확인하였다.

배양조건에 따른 생육특성 조사

배지 조성에 따른 균사생장 측정

배지 조성에 따른 균사 생장 속도를 확인하기 위해 Potato dextrose agar (PDA; BD Difco Laboratories, Detroit, USA), Malt extract & Yeast extract agar (MYA; BD Difco Laboratories), Malt extract agar (MEA; BD Difco Laboratories), Malt extract & peptone agar (MEPA; BD Difco Laboratories), Sabouraud dextrose agar (SDA; Acumedia, Michigan, USA), Modified Melin-Norkrans agar (MMNA; Bioworld, Ohio, USA)의 6가지 배지를 사용하였다. 각 배양 배지는 90 mm 직경 Petri-dish에 25 mL씩 분주하여 제작하였다. 균주 접종은 PDA 배지에서 7일 이상 키운 균주의 가장자리 부분을 5 mm 직경의 corkborer (KA-0049, Korea Ace Scientific, Seoul, Korea)를 이용해 절편화 후 접종원으로 사용하였다. 접종된 균주는 모두 25℃, 암조건의 배양기에서 배양 후 7일차에 디지털 버니어 캘리퍼스(CD-20APX, Mitutoyo, Japan)를 사용하여 균사의 직경 측정과 균사의 밀도에 대한 육안 측정을 3반복 수행하여 평균값을 산출하였다. 결과값은 측정된 균사의 직경에서 접종원의 크기를 제외한 값을 사용하였다[20-22].

온도에 따른 균사생장 측정

온도에 따른 균사 생장 속도를 확인하기 위해 20, 25, 30, 35℃의 4가지 온도조건을 선택하여 PDA 배지에서 암조건으로 7일 이상 배양하였다. 균주의 접종은 위의 방법과 동일하며 실험은 3반복으로 수행하였다.

pH에 따른 균사생장 측정

pH 조건에 따른 생장 특성을 확인하기 위해 pH 측정기(S220, Mettler Toledo, Switzerland)를 이용하여 1N-Hydrogen chloride solution (HCl)과 1N-Sodium hydroxide solution (NaOH)으로 pH 4, 5, 6, 7, 8의 다섯 가지 조건의 PDA 배지를 제조하였다. 멸균 후 pH paper로 산성도를 다시 확인하였으며, 균주의 접종은 위의 방법과 동일하게 수행하였고 25℃에서 7일 이상 배양 후 균사생장 측정을 3반복 수행하였다.

배양조건에 따른 액체배양 특성 조사

배양 기간과 배지 조성에 따른 균사 생장의 차이를 평판배지 조건과 액체배양 조건간의 비교를 위해 고체 배지와 동일한 조성에서 한천(agar)성분을 제외한 6종류의 액체배지를 1.2 L씩 제조하였다. PDA 배지에서 생육을 확인한 균주를 5 mm의 corkborer로 절편화 하여 20개의 조각을 접종한 후, 1개월과 6개월 그리고 12개월의 3가지 그룹으로 나누어 25℃에서 정치 배양하며 각 그룹의 균사체 생장을 관찰하였다. 각 그룹별로 배양이 완료된 후 filtering 과정을 거쳐 균사체와 배양여액을 분리하였고, 동결건조(PVTFD100R, Ilshinlab, Korea) 처리하여 각 그룹별 배양여액의 건조 중량을 비교하였다[20].

결과 및 고찰

배양조건에 따른 생육 특성

배지 조성에 따른 균사생장 측정

울릉도에서 채집된 표본에서 분리한 균주들은 배지에 따라 균사 생육특성에서 차이를 보였다. 먼저 Fomitiporia punctata 균주의 경우 균총의 가장자리는 대체로 흰색을, 접종원 주변부는 황갈색에 가까운 색을 띠며, 6종류의 배지 중 MEPA 배지에서 가장 빠른 균사 생장 속도를 보였지만 그 차이는 크지 않았다. MEA 배지에서는 균총의 색이 투명에 가깝고 조밀하지 않은 균사 생장을 보였다. 울릉도에서 처음 발견한 신종인 Polyporus ulleungus는 MMNA 배지에서 가장 빠르게 자랐으며 접종원 주변부로 진갈색의 질긴 균사조직을 형성하였다. Gymnopus subnudus는 균총의 표면이 편평하게 원형을 이루어 생장했으며 SDA 배지에서 균사의 밀도가 가장 높고 MEA 배지에서 가장 낮았다. Tyromyces kmetii 균사의 색은 대체로 흰색을 띠며 PDA 배지에서는 옅은 갈색의 환(環)을 형성하기도 하였으며, MYA 배지에서는 생장속도와 밀도 모두 양호한 편이었다. 마지막으로 Polyporus brumalis는 5가지 균주 중 가장 빠른 생장 속도를 보였으며, 특히 MYA와 MEPA 배지에서 가장 빠른 생육을 보였다. 그러나 MYA 배지에서는 흰색의 균사를 형성한 반면 MEPA배지에서는 갈색의 불규칙적인 균사조직을 형성하여 배지 조성에 따라 균사생장의 차이를 뚜렷하게 보여주었다. 6종류의 배지 중 Malt extract 성분이 주입된 배지의 경우 대체로 균사 생장 속도가 빠른 경향을 보였다. 그러나 Malt extract 성분 외에 Yeast extract나 Peptone과 같은 추가적인 영양원을 첨가해 주지 않았을 때 균사의 밀도가 조밀하지 않고 성기게 자라는 특징을 보였다. 균사의 생장 속도뿐만 아니라 균사의 밀도가 높게 자라는 배지를 최적 배지라고 보았을 때, MEA배지에서는 모든 균주에서 균사 밀도가 매우 얇고, 조밀하지 못하게 생장하는 것으로 보아 최적배지로의 사용은 적절하지 않은 것으로 사료된다(Fig. 2; Table 2).

온도에 따른 균사생장 측정

온도에 따른 균사 생장을 확인한 결과 대부분의 균주는 25-30℃ 사이에서 최적의 균사 생장을 보였다. 4가지 온도 범위 중 가장 고온인 35℃에서는 F. punctata, P. ulleungus, G. subnudus 균주는 전혀 생장하지 못한 반면, T. kmetiiP. brumalis는 35℃를 포함한 모든 온도에서 생장하였다. 특히 P. brumalis는 35℃ 에서도 빠른 생장 속도를 보이며 5종의 균주 중 가장 생장 온도 범위가 넓은 것을 확인할 수 있었다. P. ulleungus는 5종의 균주 중 가장 저조한 생장력을 보였으며, 30℃ 에서는 배지를 갈색으로 착색시키는 것이 확인되었다(Fig. 3; Table 3).

pH에 따른 균사생장 측정

pH에 따른 균사생장의 측정을 통해 최적 pH는 4~5인 것을 확인하였으며 약염기성인 pH 8에서는 생장 속도가 더딘 것을 확인할 수 있었다. 약염기보다는 약산성이 균사 생육에 좋은 조건이라고 제시한 기존의 연구를 뒷받침해주는 결과이다[23-25]. 5가지의 균주 중 T. kmetii는 pH 차이에 따른 균사 생장 속도의 편차가 가장 큰 것으로 나타났으며, P. brumalis는 모든 pH에서 양호한 생장을 보였다. 균사의 밀도나 형태적 특성의 차이는 다른 조건(배지조성, 온도)에 비해 크지 않았다(Fig. 4, Table 4).

배양조건에 따른 액체배양 특성

6가지 배지에서 배양기간(1개월, 6개월, 및 12개월)에 따른 배양액 상태를 비교한 결과 전체적으로 배양 기간이 길어질수록 배양액 색이 진해지는 것을 관찰할 수 있었다. 대체로 배지 자체 색상에 따라 배양액의 진하기가 달랐지만 F. punctata는 배양 기간이 길어질수록 초기 배지 색상과 무관하게 짙은 색을 띠었다. 균사체의 형태적 특징으로는 정치배양으로 인해 상층부의 매트형 생장을 보였다. 배양 속도가 빠른 P. brumalis는 배양 1개월 후부터 배양액 상층부에 균사체가 쌓여 균사 매트를 형성하였고, 6개월 차에는 F. punctata를 제외한 모든 균주가 일부 배지에서 균사 매트를 형성하였다. 배양 기간이 길어질수록 두꺼워진 균사 매트는 바닥으로 침전하였으며, 12개월 배양한 그룹의 균사체의 경우 조직이 단단하지 않고 쉽게 부서지는 모습을 보였다(Fig. 5). 종별 배양속도는 P. ulleungus가 가장 느리게, P. brumalis가 가장 빠르게 균사체가 증가하는 양상을 보였으며 이 결과는 평판배양에서의 배양속도 결과와 일치하였다.

배양여액의 건조 중량을 측정하여 배양 기간별 배지 이용률에 대한 간접적인 추정을 하고자 하였다. 배양여액의 건조 중량은 대체로 배양 기간이 길어질수록 감소하는 양상을 보였다. 특히 배양 시작부터 6개월까지는 큰 폭으로 감소하였으나, 6개월부터 12개월 사이에는 변화량이 크지 않음을 확인하였다. 그러나 T. kmetii의 SDB 배지 배양여액의 경우 초기 배지량 보다 1개월 배양여액의 건조량이 높게 측정되었으며, 이것은 동결건조 처리과정의 문제로 판단된다. 배양 초기 배지의 양과 1개월과 6개월 그리고 12개월 배양여액의 건조량을 비교하였을 때, 그 양이 점차 감소한다는 것은 균주가 배지를 지속적으로 이용하고 있다는 것을 보여준다. 그러나 접종 초기부터 6개월까지는 급격하게 감소하다가 6개월부터 12개월까지 완만한 감소폭을 보이는 것으로 보아 정치배양 조건에서의 액체배양은 1개월 이상의 배양 시간이 주어져야 배지 성분을 충분히 이용할 수 있음을 예측할 수 있다. 배양 기간별 배양여액의 건조량의 차이가 가장 큰 종은 P. ulleungus로 MEP 배지에서 배양한 시료의 경우 1개월 배양여액 건조 시료량인 7.06 g이 12개월 배양 후 0.93 g으로 약 85% 이상 감소한 것을 확인하였다. 반면 감소 폭이 가장 적은 종은 G. subnudus로 특히 ME, MEP, MMN 배지의 1개월과 12개월의 배양여액 건조량은 변화가 거의 없었다. MY 배지는 5종류의 배지 중 배양여액의 건조량 감소가 가장 큰 배지였으며 평균적으로 70%이상의 감소율을 보였다(Fig. 6).

본 연구에서는 울릉도에서 수집한 자생 버섯 균주 5종의 평판배지 배양 특성과 액체배양에서의 특성을 확인하여 자생 균류의 이용을 위한 기초 정보를 확보하였다. 5종의 야생 균주의 최적 배양온도는 25-30℃이고, pH는 4.0-5.0의 산성임을 확인하였다. 특히 P. brumalis는 35℃에서 생장속도가 가장 빠른 것으로 보아 고온성 버섯으로 판단된다. 실험에 사용한 상용 배지 중 최적 배지는 F. punctata의 경우 MEPA 배지, P. ulleungus는 MMNA 배지, G. subnudus는 MEA 배지, T. kmetii는 MMNA 배지, 마지막으로 P. brumalis는 모든 배지에서 빠른 생장 속도를 보였으며 6종류의 배지 중 MEA 배지는 모든 균주의 균사 밀도가 낮아 배양이 적합하지 않은 조성임을 확인하였다. P. brumalis는 5종의 균주 중 가장 빠른 생육속도를 보였으며, 반면에 P. ulleungus는 가장 저조한 생장을 보여 같은 속(genus)의 종이지만, 생육특성의 차이가 극명함을 보였다. 액체배양을 통해 배양 기간에 따른 배양여액의 건조 중량을 비교한 결과 배양 기간이 길수록 건조량이 감소하는 양상을 보였으며, 특히 6개월 이전까지 큰 폭으로 감소하였다. 이 결과로 정치배양 조건에서의 액체배양은 한달 이상의 배양 시간이 주어져야만 배지 성분을 충분히 이용할 수 있다는 것을 확인하였다. 본 실험의 결과로 5종의 균주에 대한 최적 배양조건을 확립하고 향후 응용 연구에 기초 자료로 활용할 수 있을 것으로 기대한다.
