Isolation of Wild Yeasts from Freshwaters and Soils in Nonsan Stream and Sapgyoho in Chungcheongnam-do, Korea, and Microbiological Characteristics of the Unrecorded Wild Yeasts

Abstract

This study aimed to isolate wild yeasts from water and soil sample of the Nonsan stream and Sapgyoho (lake) in Chungcheonnam-do, Korea, and to further characterize previously unrecorded wild yeast strains. In total, 102 strains, representing 55 different species of wild yeast were isolated from 95 samples collected from the Jangseoncheon and Ipchoncheon of Nonsan stream in Jellabuk-do and Chungcheonnam-do. Among these, 33 strains were isolated from alkalophilic yeast extract-peptone-dextrose (YPD) medium (pH 9.0), and 9 strains were isolated concurrently on general YPD medium (pH 6.5) and alkalophilic medium. Seventeen strains of Cryptococcus laurentii were predominantly isolated. Additionally, 65 strains, representing 27 different species of wild yeast were isolated from 58 samples obtained from Sapgyoho (lake). Among the 82 isolated wild yeast strains, 8 strains, including Candida fructus JSC 72-1(JSL-GGU 015), had not previously been recorded. All 8 previously unrecorded yeasts were oval in shape except C. fructus JSC72-1(JSL-GGU-015), and only the Filobasidium chernovii JSC39-1(JSL-GGU-013) strain formed spores. All strains except Pseudosydowia eucalypti JSC23-6(JSL-GGU-012) grew well in yeast extract-peptone-dextrose (YPD) and yeast extract-malt extract media and grew in vitamin-free medium. Four strains, including P.eucalypti JSC23-6(JSL-GGU-012) grew well in 15% NaCl-containing YPD medium. F.chernovii JSC39-1(JSL-GGU-013) and Sirobasidium intermedium JSC7-3(JSL-GGU-014) assimilated lactose, and five strains, including F. chernovii JSC39-1(JSL-GGU-013) also assimilated starch. All strains were resistant to 800 ppm of Ca, Cu, Li, and Mg ions.

서론

진균류인 효모는 오래전부터 전통 발효식품 제조 등에 이용되어 왔고[1-3] Sacccharomyces 속 균을 중심으로 외래 유전자 발현을 위한 분자생물학적 연구 재료 균주로 귀중하게 이용되어오고 있다[4]. 또한, 비타민과 아미노산 및 일부 효모들의 여러가지 생리활성 물질들[5-8]과 probiotics균들이 보고 되고 있다[9].

지금까지 효모균의 분리 및 동정은 주로 전통 발효식품과 이들의 원료로부터 실시되어 왔고[1,2] 최근에는 야생화와 토양 등 자연환경으로부터 다양한 야생효모들이 분리, 동정되어 이들의 효모 종 분포특성 [10-21]과 비병원성 균들에 대한 생리활성 물질 생산 등이 보고되었다[7,8,12,16,17]. 또한, 담수환경으로부터의 새로운 효모균류 발굴 목적으로 필자 등은 낙동강과 영산강, 금강 등 우리나라 주요 강과 대전천 등 하천 주변 물과 토양 등으로부터 다양한 야생 효모들을 분리, 동정 하여 보고하였고[22-27] 이들 야생화와 토양 및 담수 환경으로부터 150여종의 국내 미기록 야생효모들을 선별하여 이들의 균학적 특성 등을 보고 하였다[18,22,25,26,29-31].

한편, 금강 하류의 제1지류인 논산천은 전북 완주군 외사봉줄기에서 발원하여 귀목동천과 안심천, 장선천, 입촌천, 웅천 등을 거쳐 논산 저수지(탑정호수)로 유입된 후 백마강과 강경을 거쳐 서해로 흘러간다[32-33]. 이들 하천은 대체로 수량이 풍부하고 모래와 잔자갈이 많고 논과 밭이 많으며 벼, 딸기, 양파, 고구마 등이 주로 경작되고 있다. 삽교호는 아산만의 삽교천 하구를 가로 막은 인공 담수호로서 충남 당진과 아산, 예산, 홍성 등의 무한천, 삽교천, 곡류천 등의 물들이 유입되고 이들 지역의 넓은 평야와 간석지 등의 농업용수 확보를 목적으로 만들어졌다. 그러나, 이렇게 농업용수와 생활환경 조정자로서 중요한 역할을 하는 논산천과 삽교호에 대한 이들 지역의 생태 환경 연구는 매우 미흡하여 Lee 등[33]이 논산천의 어류군집 분포 특성이 보고되었을 뿐이다.

따라서 본 연구에서는 금강 하류의 지천으로 전북 완주와 충남 논산으로 이어지는 논산천과 충남 아산만의 삽교호 주변의 야생 효모 종 분포특성을 알아보기 위해 이들 지역 주변의 물과 토양 등으로부터 야생 효모들을 분리하여 동정하였다. 또한, 이들 야생 효모들 중 국내에 보고되지 않은 미기록 야생 효모들을 선별하여 이들의 균학적 특성을 조사하였다.

재료 및 방법

야생 효모의 분리 및 동정

금강 하류의 지천인 논산천을 이루는 전북 완주군 장선천(36°04'50.0"N, 127°17'05.7"E), 충남 논산의 입촌천(36°08'22.5"N, 127°14'01.5"E)과 충남 아산만의 삽교호(36°52'16.9"N, 126°51'34.2"E)주변 물, 토양과 야생화 등 시료 153점들을 2021년 3월과 5월, 7월에 채취하였다.

이들 시료들을 멸균 튜브에 넣고, 5 mL의 멸균수를 첨가한 후 2시간 동안 진탕하였다. 이들 현탁액 일부를 스트렙토마이신(100 µg/mL)과 앰피실린(100 µg/mL)이 들어 있는 yeast extract-peptone-dextrose (YPD, pH 6.5) 한천배지에 도말하고 30℃에서 48시간 배양한 후 형성된 효모 집락들을 일반 야생 효모균주로 분리하였고[31] YPD배지의 초기 pH를 9.0으로하여 분리한 효모들을 호알칼리성 효모로 분리하였다.

분리 효모들의 동정을 위하여 먼저 분리 균주들의 26S rDNA의 D1/D2 부위를 colony PCR과 전기영동을 실시하여 효모 밴드를 확인한 후 이들의 염기서열들을 결정하고 결정된 염기서열들을 NCBI의 BLAST를 사용하여 데이터베이스에 등록되어 있는 효모들과의 상동성 비교와 분자생물학적 유연관계를 분석하여 동정하였다[25-31].

국내 미기록 효모들의 선별 및 특성

위와 같이 분리, 동정한 야생 효모들을 대상으로 국립 생물자원관 데이터 베이스와 한국 진균 관련 학술자료들을 이용하여 국내 미기록 효모들을 선별한 후 일반 미생물 실험방법 등을 이용하여 이들의 형태학적 특성과 배양학적 특성을 조사하였고[25,31], 탄소원들에 대한 자화성과 발효성은 yeast nitrogen base (YNB) 배지를 최소배지로 하여 각각의 당과 당알코올 등을 1%씩 첨가하고 Durham tube를 넣어 30℃에서 72시간 배양한 후 생육과 이산화탄소 생성 유무를 확인하여 조사하였다.

또한, YPD배지에 에탄올을 0-10%, CoCl2등의 중금속들을 400/800 ppm씩 각각 첨가하여 30℃에서 72시간 배양한 후 이들의 생육도를 측정하여 내성을 조사하였다[1,2].

결과 및 고찰

충남 논산천 주변 생태환경으로부터 야생 효모의 분리 및 미기록 야생 효모들의 선별

논산천을 이루는 전북 장선천과 충남 입촌천 주변의 물과 토양 95점에서 일반 효모 69균주와 호알칼리성 균주 33주 등 모두 55종, 101균주의 야생 효모들을 분리하였고 이들 야생효모 종에서 9종의 효모들은 일반 효모 배지와 알칼리성 배지에서 동시에 분리되었다(Table 1). 또한, 이들 101균주들 중 Cryptococcus laurentii 균주가 17균주로 가장 많았고 Sporobolomyces carnicollor 균주도 7균주로 많이 분리되었다.

한편, Candida panamericana JSC6-3(JSL-GGU-011), Pseudosydowia eucalypti JSC23-6(JSL-GGU-012), Filobasidium chernovii JSC39-1(JSL-GGU-013), Sirobasidium intermedium JSC7-3(JSL-GGU-014), Candida fructus JSC72-1(JSL-GGU-015), Candida stauntonica JSC20-1(JSL-GGU-016) 등 6균주들이 우리나라에 아직까지 보고되지 않은 국내 미기록 야생효모들로 선별되었다(Table 1).

충남 아산 삽교호 주변 생태환경으로부터 야생효모의 분리 및 동정

충남 아산만의 삽교호 주변 물과 식물체 부식물 및 토양 58점에서 YPD (pH 6.5) 배지에서 34균주, 호알칼리성 YPD (pH 9.0) 배지에서 31균주 등 모두 65균주의 야생효모들이 분리되었다(Table 2). 이들 65균주 중 9균주가 일반 효모 배지와 호알칼리성 배지에서 공통적으로 분리되었다. 이들 중 Candida tritomae 균주가 전체 19%인 12균주로 가장 많았고 Pseudozyma aphidis 균주도 6균주로 많이 분리되었다.

한편, C. tritomae SG 55M-3(JSL-KSS-003) 균주와 Kwoniella mangrovensis ASG 21W-2(JSL-KSS-004) 균주들이 국내 미기록 효모들로 최종 선별되었다(Table 2).

최근 담수환경으로부터 새로운 야생균류 발굴에 대한 관심이 높아지고 있고 이들 환경의 효모 종 분포 특성을 조사하기 위해 필자 등은 금강 하류 충남 서천부터 중류인 대전광역시 신탄진의 대청호 주변과 금강 상류인 금산 지역의 담수와 토양 환경에서 Candida subhashii, Bullera japonica, Cry. magnus, C. chauliodes, Holtermanniella takashimae, Cryptococcus, Rhodotonular 속 균들이 많이 분포하고 있음을 보고하였다[24,25,27]. 또한 낙동강과 영산강[22], 대전천과 갑천[23] 등지에서 다양한 야생 효모들이 분리, 동정되었고 이들 중 국내 미기록 효모들의 특성 등이 보고되었다[22,26,27]. 한편, 전 등[26]은 태백 하천에서 국내 미기록 야생 효모들을 분리하여 이들의 특성을 보고한 바 있다.

국내 미기록 야생 효모들의 균학적 특성 및 효소활성

위와 같이 금강의 지천인 논산천과 서해안 아산만의 담수 호수인 삽교호 주변에서 분리한 야생 효모들중 국내 미기록 야생 효모들로 최종 선별된 C. panamericana JSC6-3 (JSL-GGU-011), P. eucalypti JSC23-6 (JSL-GGU-012), F. chernovii JSC39-1 (JSL-GGU-013), Sir. intermedium JSC7-3 (JSL-GGU-014), C. fructus JSC72-1 (JSL-GGU-015), C. stauntonica JSC20-1 (JSL-GGU-016, C. tritomae SG 55M-3 (JSL-KSS-003) 균주와 K. mangrovensis ASG 21W-2 (JSL-KSS-004) 등 8주들에 대하여 균학적 특성으로 형태학적, 배양학적 특성과 중금속 등에 대한 내성 및 탄소원들에 대한 자화성과 발효성을 조사하였다 (Table 3, 4),

형태학적, 배양학적 특성; 이들 미기록 효모들의 형태적, 배양적 특성과 이들의 phylogenetic trees는 각각 Table 3, Fig. 1-A, Fig. 1-B와 같다. 이들 미기록 효모들 중 C. fructus JSC72-1외에는 모두 난형으로 출아에 의해 영양 증식을 하였고 F. chernovii JSC39-1 균주만이 포자를 형성하였으며 모든 균주들이 의균사를 형성하지 않았다. P. eucalypti JSC23-6 균주만이 vitamin-free배지에서 생육하지 못했고 대부분의 균주들이 40% 포도당을 함유한 YPD배지에서 생육하는 내당성을 보였으며 특히 C. panamericana JSC6-3등 3균주들은 15% NaCl을 함유한 YPD배지에서도 생육이 비교적 양호한 호염성균으로 내염성 대사산물 생산에 매우 유용할 것으로 사료된다.

중금속 이온들에 대한 내성; 8주의 국내 미기록 야생 효모들의 중금속 이온들에 대한 내성을 조사한 결과 모든 미기록 효모들이 Ca, Cu, Li, Mg 이온들에 대하여 800 ppm까지 내성을 보였고 특히 삽교호에서 선별한 C. tritomae SG55M-3 균주와 K. mangrovensis ASG 21W-2 균주들은 Ca, Cu, Li, Mg, Co, Fe, Zn 이온들에 대하여 800 ppm까지 강한 내성을 보였다. 이러한 중금속 이온들에 대한 내성은 이 등[1]이 우리나라 전통 장류에서 분리한 Saccharomyces 속 균들과 호염성 Zygosaccharomyces 속 균들이 Pb, Hg, Co, Cu 등의 2가 금속이온들에 대하여 800 ppm 내성을 보였다는 결과와 유사하였다.

탄소원들에 대한 자화성과 발효성; 미기록 야생 효모들의 탄소원으로 당류와 당알코올 및 유기산들에 대한 자화성과 발효성을 조사한 결과 Table 4와 같다. 미기록 효모들 모두 포도당과 fructose, sucrose들을 자화시켰고 특히 F. chernovii JSC39-1등 3균주들은 lactose를 자화시켰다. C. panamericana JSC6-3등 5균주들은 전분을 자화시켰고 모든 미기록 효모 균주들이 구연산을 이용하였으나 초산은 이용하지 못하였다.

한편, 이들 국내 미기록 효모들에 대한 국외 연구로 Suh 등은 버섯 자실체 서식 딱정벌레의 소화기관 등에서 C. panamericana [34]와 C. tritomae 균주를 처음 분리하여 보고하였고, Khan 등은 소아암 환자의 코 조직에서 F. chernovii를 분리하여 보고하였다 [35, 36]. 또한 Baraniak 등은[37] xylose를 자화시키는 C. fructus를 선별하여 2%의 xylose 발효배지에 리터당 1g 이상의 ethanol을 생산하였다고 보고하였고 EI-Gendy 등[38]은 알긴산으로 고정화시킨 C. stauntonica로부터 리터당 4.4g의 bioethanol을 생산하였음을 보고하였다 [38].
